摘要
目的 探讨P-p38MAPK及TNF-α在子痫前期中的发病机制.方法 取2016年8月至2018年6月单胎足月阴道分娩孕妇57例为观察对象,其中正常足月组20例,轻度子痫前期18例,重度子痫前期19例,抽取相应孕妇住院分娩时未进行任何药物治疗前肘静脉血4~5ml后分离血浆,并获得相应患者胎盘组织.采用ELISA检测孕妇血浆中TNF-α浓度,并采用常规SABC法检测57例孕妇胎盘组织中P-p38MAPK表达的强度.结果 正常孕妇及轻、重度子痫前期患者血浆TNF-α水平依次为(39.27±1.16)pg/ml、(325.81±11.58)pg/ml及(635.24±32.82)pg/ml;正常孕妇及轻、重度子痫前期患者胎盘组织中P-p38MAPK灰度值依次为(13.11±1.07)、(29.24±1.14)及(43.17±6.21);与正常妊娠孕妇相比,轻、重度子痫前期患者血浆TNF-α水平依次升高,差异均具有显著性.胎盘组织内P-p38MAPK表达水平亦呈相类似的变化.结论 孕妇体内TNF-α及P-p38MAPK参与了子痫前期的发病,并作为预测子痫前期严重程度中有一定的应用前景.
Objective To investigate the pathogenesis of p-p38MAPK and TNF-αin preeclampsia.Methods A total of 57 pregnant women from August 2016 to June 2018 with full term singletons born by normal vaginal delivery were studied.There are 20 cases of term pregnancy and 37 cases of pregnancy complicated with preeclampsia(18 cases of mild and 19 cases of severe preeclampsia).When the corresponding pregnant women delivery in hospital without any drug treatment,4 to 5 ml blood of the elbow vein was separated and the placental tissue of the corresponding patient was obtained.ELISA was used to detect the plasma concentrations of TNF-αin the 57 pregnant women,and the expression of P-p38MAPK was studied by immunohistochemistry SABC method in corresponding placental tissue.Results The plasma levels of normal pregnant women and patients with mild and severe preeclampsia were(39.27±1.16)pg/ml,(325.81±11.58)pg/mland(635.24±32.82)pg/ml.The P-p38 MAPK grayscale values in placental tissues of normal pregnant women and preeclampsia patients were(13.11±1.07),(29.24±1.14)and(43.17±6.21).Compared with normal pregnant women,the plasma levels of TNF-αin preeclampsia patients increased and there were significant differences.The level of P-p38MAPK expression in placenta tissue also showed similar changes.Conclusion TNF-αand P-p38 MAPK are involved in the pathogenesis of preeclampsia in pregnant women,and may be used as predictors of disease severity in preeclampsia.
