莫沙必利对心理应激诱导小鼠食管炎症发生的影响及作用

摘要 目的探讨莫沙必利对心理应激小鼠食管炎症发生的影响及作用。方法8周龄,SPF级雄性昆明小鼠随机分为3组,即正常对照组(6只)和慢性束缚应激组(心理应激组,6只)以及莫沙必利干预组(6只)。心理应激组及莫沙必利干预组小鼠采用限制活动的自制束缚器,小鼠入束缚器内限制其活动1天1次(1次2 h),实验持续进行14天;采用RT-PCR方法对比分析3组小鼠食管组织氧化应激,抗氧化蛋白以及炎性因子的mRNA表达水平。结果莫沙必利显著抑制心理应激诱导小鼠食管组织炎症反应发生。心理应激诱导小鼠食管组织氧化应激指标(Nox-4)mRNA水平的高表达(P<0.01);同时,心理应激降低其抗氧化蛋白指标(Nrf-2、HO-1)的mRNA表达水平(P<0.01);莫沙必利干预能够有效改善心理应激诱导氧化应激(Nox-4)和抗氧化蛋白(Nrf-2、HO-1)的异常表达(P<0.05)。心理应激诱导小鼠食管组织炎性因子(IL-1β、TNF-ɑ)的mRNA水平显著增高(P<0.01);莫沙必利干预能够抑制心理应激诱导食管组织炎性因子的高表达(P<0.01)。结论莫沙必利干预有效抑制心理应激小鼠食管炎症、氧化应激以及炎性因子的异常表达。 ObjectiveTo investigate the effect of Mosapride on esophageal inflamm ation in mice under stress.MethodsEight week old SPF male Kunming mice were randomly divided into three groups:normal control group(n=6),chronic restraint stress group(stress,n=6)and Mosapride intervention group(n=6).The mice in the stress group and Mosapride intervention group were restricted in the self-made restraint device for 2 h every day,and that restraint experiment kept 2 h/each day of 14 consecutive days.The mRNA expression levels of oxidative stress,antioxidant protein and inflammatory factors in esophageal tissues of three groups of mice were detected and analyzed by RT-PCR method.ResultsMosapride significantly inhibited the occurrence of esophageal inflammation induced by stress in mice.Stress significantly increased the mRNA levels of oxidative stress marker(Nox-4)in esophageal tissue(P<0.01);At the same time,stress significantly reduced the mRNA expression levels of antioxidant proteins(Nrf-2,HO-1)(P<0.01);Mosapride intervention can effectively improved the stress induced above abnormal expressions of oxidative stress and antioxidant proteins(P<0.05).Stress significantly increased the mRNA expression levels of inflammatory factors(IL-1βand TNF-ɑ)in esophageal tissue(P<0.01);Mosapride intervention could inhibit the stress induced high expressions of these inflammatory factors in esophageal tissue(P<0.01).ConclusionMosapride intervention can effectively inhibits esophageal inflammation,oxidative stress and abnormal expression of inflammatory factors in mice under stress.
