摘要
用野生型发根农杆菌A4和R1601菌株转化蒙古黄芪试管苗的不同部位,建立了发根转化体的培养系,硅胶薄层层析显示转化组织中含有冠瘿碱.试验不同的转化条件和理化因子对提高转化率的影响.结果表明,R1601的转化频率高于A4,外植物中下胚轴的转化频率最高,基本培养基以MS转化效果最佳,外植体预培养3 d、与农杆菌共培养4 d、培养基中附加100μmol/L的乙酰丁香酮、0.3 mg/L IBA的条件下,诱导率最高达42.2%.
The pieces of sterile seeding of Astragalus membranaceus Bunge Var.mengholicus(Bunge) Hsiao were infected with [WTBX]Agrobacterium rhizogenes strains A_4 and R_(1601).The transformants(hairy roots and transformed calluses) were obtained by selection on hormone-free medium.The transformed root cutures were assayed by TLC,with positive result for opines.The effect of several factors on transformation frequency was studied.The results indicated that Strain R_(1601) has higher transformation frequency than that...
