摘要
以Novozym4 35脂肪酶为催化剂 ,正己烷为有机溶剂 ,丙二醇为乳化剂 ,酶促合成葡萄糖硬脂酸酯。考察了脂肪酶加入量、反应温度、反应时间、底物配比、体系含水量、振荡速率对酶促酯化反应的影响 ,确定了酶促反应的工艺条件 ;同时对产物进行了定性、定量分析。研究结果表明 :酶加入量为脂肪酸质量的 10 % ,底物酸、糖的摩尔比为 1∶1,温度为 6 0℃ ,含水量为有机溶剂的5 3% ,反应时间为 4 .5h ,振荡速率为 10 0r/min时 ,硬脂酸最大转化率可达 93.4 0 %。
The synthetic reaction of glucose stearic acid ester and its influence factos were discussed.This reaction used the Novozym435 lipase as catalyst, the hexane as solvent and the propylene glycol as emulsifier.The influence of the lipase concentration, the temperature, the reaction time, the mol ratio of stearic acid to glucose, the moisture content and the speed of shaking bed on the rate of conversion were also researched. The determination of products was also described. The resuls indicate that when the lipase concentration is 10% of the stearic acid, the mol ratio of substrate stearic acid to glucose is 1∶1, the reaction temperature is 60℃,the moisture content is 5.3% of solvent,the reaction time is 4.5 hours and the speed of shaking bed is 100 r/min, the conversion of stearic acid can reach 93.4%.
