重度子痫前期胎盘细胞凋亡及其基因的表达 被引量:1

Cell apoptosis and expression of apoptosis-associated genes in placenta of patients with severe pre-eclampsia

摘要 目的探讨重度子痫前期患者胎盘绒毛滋养细胞的凋亡情况及凋亡相关基因的表达。方法选择重度子痫前期患者及正常以社会因素剖宫产者各12例,以TUNEL法检测胎盘绒毛滋养细胞的凋亡率,免疫组化法检测Bcl-2、Bax和Fas的表达。结果子痫前期患者胎盘绒毛滋养细胞的凋亡率明显高于对照组(F=22.65,P<0.05);与正常对照组比较,Bax和Fas在重度子痫前期组胎盘绒毛滋养细胞的表达水平显著增加,差异有显著性(χ2分别为4.1958、6.1714,均P<0.05);Bcl-2在两组胎盘绒毛滋养细胞的表达无明显差异性。结论子痫前期患者胎盘绒毛滋养细胞的凋亡率增加及促凋亡基因Bax和Fas的高表达导致胎盘缺血和缺氧,可能与重度子痫前期的病因及病理过程有关。 Objective To investigate cell apoptosis and expression of apoptosis-associated genes in trophocytes of placenta of patients with severe pre-eclampsia.Methods 12 women with severe pre-eclampsia,and 12 normal pregnant women underwent cesarean section due to social factors(as controls) were selected.TUNEL was used to detect apoptosis rate and immunohistochemical method was used to detect expressions of Bcl-2,Bax and Fas in the trophocytes of placenta.Results The apoptosis rate in the trophocytes of placenta of patients with severe pre-eclampsia was significantly higher than that in the control group(F=22.65,P<0.05).The expression levels of Bax and Fas in the trophocytes of placenta of patients with severe pre-eclampsia were higher as compared with the control group and there were significant differences(χ2=4.1958,6.1714,respectively,Both P<0.05).While in expression of Bcl-2 in the trophocytes of placenta,there was no significant difference between the pre-eclampsia group and the control group.ConclusionThe apoptosis rate in trophocytes of placenta of patients with severe pre-eclampsia was increased.Over-expressions of apoptosis-promoting genes Bax and Fas in the trophocytes cause hypoxia and ischemia of placenta,which may be associated with occurrence and development of severe pre-eclampsia.
