热疗联合化疗及放疗对人结肠癌裸鼠移植瘤E-cad与β-cat表达影响的研究 被引量:3

摘要 目的:探讨热疗、化疗、放疗、热化疗、热放疗及热放化疗对人结肠癌裸鼠移植瘤上皮细胞黏附因子表达的影响。方法:将人结肠癌细胞株HT-29移植在裸鼠后肢,于实验室模拟大肠癌临床热疗、化疗及放疗方案,对荷瘤裸鼠分别实施热疗、化疗、放疗、热化疗、热放疗及热放化疗。分别于治疗前和治疗后2、4、8、12、24、48和72h,处死裸鼠,利用免疫组化染色观察肿瘤细胞膜、细胞质及细胞核钙黏附蛋白(E-cad)及β-链蛋白(β-cat)表达的形态学变化。结果:治疗后8~72h,6种治疗方法均能不同程度上调人结肠癌细胞株HT-29裸鼠移植瘤中E-cad的表达(P<0.05),而对β-cat表达的影响均不明显,P>0.05;治疗后12~72h,E-cad在热化组、热放组及热放化组的表达明显高于热疗组、化疗组及放疗组。结论:热疗可能通过增强放化疗对人结肠癌细胞株HT-29裸鼠移植瘤中E-cad的表达的改变,发挥对放化疗的增敏作用。 OBJECTIVE:To investigate the changes in E-cadherin(E-cad)and β-catenin(β-cat)expressions after hyperthermia,chemotherapy and radiotherapy in human colon cancer cells transplanted in nude mice.METHODS:Human colon cancer cells(HT29)were transplanted into the behind limbs of naked mice.Under the laboratory simulated clinical prescription of hyperthermia,chemotherapy and radiotherapy,nude mice bearing human colon carcinoma underwent hyperthermia,chemotherapy,radiotherapy,chemohyperthermia,radiohyperthermia and ...
