摘要
目的 :观察兔骨髓基质细胞经bFGF刺激后 ,其VEGF因子表达及细胞生物行为的变化 .方法 :取兔双侧股骨骨髓基质细胞 ,采用骨髓基质细胞体外培养技术 ,分别以不同剂量bFGF刺激细胞 ,并设立空白对照组 .培养 5d后 ,进行细胞形态 (HE染色 )、增殖情况 (细胞记数法与MTT法 )、碱性磷酸酶 (改良钙钴法染色 )、成骨结节 (图像分析 )、VEGF阳性细胞率 (免疫组化染色 )等项目的检测 .结果 :不同剂量组间各项观测指标差异均显著 ,结果均优于空白对照组 .bFGF促进细胞增殖与VEGF表达最佳剂量为 12 0 0kU·L 1.结论 :bFGF能促进骨髓基质细胞增殖 ,促进VEGF的表达 ,其剂量与效果间存在明显相关关系 .bFGF为 12 0 0kU·L-1时 ,促进骨髓基质细胞表达VEGF及促进细胞增殖作用同时达到最佳效果 .
AIM: To investigate the effects of basic fibroblast growth factor (bFGF) on vascular endothelial growth factor (VEGF) expression and the biological behavior of marrow stromal cells. METHODS: The marrow stromal cells of the femur of rabbits were procured and cultured. The samples were treated with different doses of bFGF and the control was not treated. Five days later, the effects were observed by histopathological analysis, cells multiplication, alkaline phosphatase activities detection, the computer analysis of bone clusters and the detection of ratio of VEGF positive cells. RESULTS: There was statistically significant difference between different dose groups in all the detection items and the results of treatment groups were all better than that of the control group. CONCLUSION: bFGF can stimulate both the multiplication and osteogenic potential of marrow stromal cells in a dose dependent manner and the best dose of bFGF is 1200 kU·L -1 .
