摘要 目的 :观察腺病毒载体转染兔心肌细胞的有效性和外源性基因表达的时相性。 方法 :用 Adv5 - CAG粘粒(对照组 )或 Adv5 - CAG/L ac Z质粒 (治疗组 )转染兔心肌 ,转染后第 3、7、14、2 8天取被转染心肌组织行 X- gal染色及β-半乳糖苷酶活性检测。 结果 :X- gal染色示 ,治疗组转染后第 3、7、14、2 8天注射部位心肌均见蓝染 ,尤以第 7、14天明显 ,而对照组所有心肌均无蓝染 ;β-半乳糖苷酶活性检查示治疗组转染后第 3天心肌中即有β-半乳糖苷酶表达 ,第 7、14天表达达高峰 ,2 8天表达量明显减少。 结论 :重组腺病毒载体可成功转染兔心肌细胞 ,外源性基因在兔心肌细胞中能有效地表达并能维持 Objections: The efficiency and stability of adenovirus mediated gene transfer into rabbit myocardium were investigated. Methods: Adv5 CAG (control group) or Adv5 CAG / LacZ (treatment group) was directly injected into myocardium via a thoracic approach in adult rabbits. On the 3rd,7th,14th and 28th day after injection, the myocardium of the two groups were explored. All samples were analyzed for the presence of β galactosidase activity and histological staining. Results: Microscopic examination of section from hearts of the control group revealed no X gal staining. The treatment group revealed blue staining. Hearts infected with Adv5 CAG/ LacZ showed significently increased β gal enzymatic activity compared with hearts infected with Adv5 CAG.β galactosidase activity was not detected in the hearts of the control group. Conclusions: Adv5 vectors provide an efficient method for in vivo expression in myocardium. The high level of gene expression only persisted about one month.
