摘要
目的:研究肺癌细胞表面Fas表达水平与抗Fas治疗生物学效应之间的关系。方法:将低表达Fas的肺癌细胞A-549细胞通过脂质体介导的方法转染Fas基因,并用流式细胞直接免疫荧光术检测Fas的表达;采用鼠抗人Fas单克隆抗体DX-IgG1κ诱导转染Fas基因的和未转染Fas的A-549细胞的细胞凋亡作用,并用碘化丙啶染色流式细胞术检测。结果:转染Fas基因后的A-549细胞Fas分子表达明显增加,Fas分子表达率从26.11%增加到68.69%;鼠抗人Fas单克隆抗体DX-IgG1κ诱导的转Fas基因的A-549细胞凋亡数明显增多,达66.3%,而未转基因的A-549细胞凋亡较少,仅7.4%。结论:人类恶性肿瘤细胞系对Fas抗体触发凋亡信号的敏感性主要依赖于细胞表面Fas的表达水平。
Objective:To investigate the relationship between expression level of Fas and anti-Fas biological responsiveness.
