摘要
考察了双亲灭活条件、聚乙二醇(PEG)相对分子质量、PEG浓度、融合时间、融合温度及无机离子浓度对褐黄孢链霉菌(Streptomyces gilvosporeus)SG-2002-1和SG-2002-2原生质体融合率的影响。分别采用紫外灭活和热灭活法,灭活条件为紫外灭活2 min,热灭活为60℃水浴1 h。原生质体融合最佳条件为:50%的PEG 6000,融合温度25℃,融合时间为8 min,CaCl2浓度为0.05 mol/L,MgCl2浓度为0.02 mol/L,在此条件下融合率可达5.0%。
The important factors including biparental inactivation,PEG relative molecular weight,PEG concentration,the fusion time,the fusion temperature,the concentration of inorganic ion are studied for protoplast fusion of S.gilvosporeus SG-2002-1 and S.gilvosporeus SG-2002-2.Protoplasts are completely inactivated by UV for 2 min and heat for 1 h at 60℃,respectively.The optimum conditions of protoplast fusion are as follows:PEG relative molecular weight of 6 000,PEG concentration of 50%,the fusion time of 8 min,the...
