慢性碘过量增加大鼠甲状腺细胞fas、fasL和TRAIL蛋白的表达 被引量:2

摘要 目的探讨慢性轻中度碘过量对Wistar大鼠甲状腺细胞fas、fasL、TRAIL、bcl-2和bax蛋白表达的影响,观察限碘后的恢复情况。方法将大鼠分入4组:对照组(N组)、1.5倍组、3倍组和6倍组,每日给碘量分别为4、6、12和24μg,于实验1~8个月内分批处死动物。部分8个月组大鼠以对照组条件饲养3个月后再处死。应用流式细胞术和免疫组织化学方法检测fas、fasL、TRAIL的表达,采用免疫组织化学方法检测bcl-2和bax蛋白的表达。采用砷铈催化分光光度法测定尿碘浓度。结果碘过量8个月时,甲状腺细胞fas、fasL、TRAIL蛋白表达阳性率随给碘剂量的增加,显著增加,与给碘量显著正相关,限碘后不恢复。碘不影响bcl-2和bax蛋白的表达。结论慢性碘过量增加大鼠甲状腺细胞fas、fasL和TRAIL蛋白的表达。 ObjectiveTo investigate the chronic effects of mild and moderate iodine excess and iodine restriction on expressions of apoptosis-related proteins of rat thyroid. MethodsWistar rats were exposed to 4 different doses of iodine:4 μg/d(N group) ,6 μg/L(1.5-fold group) ,12 μg/L(3-fold group) and 24 μg/L(6-fold group) for 1,2,4 and 8 months.
