复方积雪草2号治疗IgA肾病大鼠的实验研究 被引量:16

摘要 目的:观察复方积雪草2号对IgA肾病(IgAN)模型大鼠蛋白尿、血尿及肾脏病理改变的改善情况。方法:复合免疫法制造大鼠IgAN模型,分设正常对照,模型,缬沙坦,复方积雪草2号高剂量、中剂量、低剂量6组。观察各组大鼠尿红细胞、尿蛋白及肾组织病理变化。结果:①模型组血尿、蛋白尿较正常组明显增多,各治疗组的血尿、蛋白尿均较模型组改善(P<0.05),其中复方积雪草2号改善血尿作用优于缬沙坦(P<0.05)。②免疫荧光镜下观察模型组大鼠肾小球系膜区有较强IgA荧光,光镜下系膜基底膜面密度明显增加,而毛细血管袢面密度明显减少;各治疗组系膜区内的IgA荧光强度、系膜增生情况均较模型组明显改善(P<0.05)。③电镜下模型组系膜细胞增生,基质明显增多,有足突融合,各治疗组足突融合情况均较模型组明显改善(P<0.05)。结论:复方积雪草2号能降低IgAN大鼠肾组织内IgA的沉积、减少系膜增生、减轻足细胞融合,从而改善IgAN大鼠的肾脏损害,减少蛋白尿、血尿。 Objective:To observe the effects of compound jixuecao Ⅱ on proteinuria, hematuria and renal pathology of rats with IgA nephropathy. Methods: Models with IgA nephropathy were induced by multiplex immunity. 48 rats were divided into 6 groups, normal, model, western medicne, jixuecao Ⅱ (low, middle, high) three dose groups, hematuria, proteinuria were detected before and after treated, renal pathologic changes of each group and immuno-fluorescent were examined by microscope. Results:Hematuria and proteinuria in each treated group were relieved obviously, the improving effect of jixuecao Ⅱ on hematuria was better than that of western medicine. IgA density of fluoroscope was stronger in model rats, area density of basilar membrane increased obviously, area density of glomerular capillary loops decreased significantly, all these were improved in each treated group obviously. Mesangioproliferation, electron-dense deposition and podocyte fusion were observed in model rats by electro-microscopy, while these changes in each treated group were not remarkblely. Conclusion:Compound jixuecao II can improve kidney injury, relieve hematuria and proteinuria, Which is related to lower IgA depositing in medangial area, reduce mesangioproliferation and ameliorate podocyte fusion.
