摘要
目的:探讨血管平滑肌肌动蛋白α(vascular smooth muscle α-actin,VSMA-α)表达变化在移植心脏血管病变及纤维化中的作用。方法:建立大鼠心脏移植模型,并将实验大鼠分为急性排斥组、慢性排斥组、同系移植组、正常心脏组。采用Masson和Van Gieson染色观察心肌的纤维化,计算血管狭窄的指数。免疫组织化学染色分析VSMA-α表达变化与心肌纤维化和血管病变的关系。结果:正常心脏组、同系移植组、急性排斥组、慢性排斥组心肌纤维化评分分别为0.00±0.00,0.63±0.52,1.75±0.71和2.75±0.46,慢性排斥组心肌纤维化明显高于正常心脏及同系移植心脏(均P<0.01)。而冠状动脉狭窄指数分别为0.08±0.02,0.09±0.04,0.12±0.05和62.86±17.18,其中慢性排斥组移植心脏血管较其它组明显狭窄(均P<0.01)。慢性排斥组移植心脏的心肌细胞大量表达VSMA-α。结论:血管平滑肌肌动蛋白α基因的重新激活及其蛋白的异位表达与移植心脏血管病变及心肌纤维化有关。
