摘要
目的 观察卵巢癌抗独特型单链抗体 6B11ScFv与鼠粒细胞巨噬细胞集落刺激因于(murine granulocvte-macrophage colony-stimulating factor.mGM-CSF)的融合蛋白(6B11mGM)在小鼠体内诱导抗肿瘤免疫反应情况,探讨此种融合蛋白作为卵巢癌疫苗的可能性及适当的免疫方法。方法 用融合蛋白6B11mGM免疫BALB/c小鼠,制备抗血清。采用间接ELISA及免疫流式方法分析抗血清特征。结果 经ELISA检测,融合蛋白6B11mGM免疫小鼠后,可诱导机体产生AB3。同时可刺激小鼠脾脏淋巴细胞CD4+细胞明显升高,CD8+细胞略有升高。结论 融合蛋白6B11mGM可诱导机体产生特异体液免疫和细胞免疫反应,为抗独特型卵巢癌疫苗用于临床奠定了基础。
Objective To observe whether antitumor immune response c an be induced in BALB/c mice immunized with a fusion protein(6B11mGM) of anti-idiotypic single chain antibody (6B11scFv) and murine granulocyte-macrophage colony-stimulating factor (GM-CSF) instead of ovarian carcinoma antigen.Methods BALB/c mice were immuni zed repeatedly with 6B11mGM.Indirect ELISA test and immune flow cytometer were used for analyzing the characterization of anti-idiotypic ScFv(Ab3).Result s Ab3 could be detected in the sera of immunized mice with 6B11mGM by ELISA test .Moreover,it stimulated CD4+T cell from the spleen of BALB/c mice to proliferate and CD8+ T cell to proliferate slightly.Conclusion 6B11mGM probably induce b oth humoral and cellular immunity against ovarian carcinoma in vivo .It illus tra ted a strong basic for using fusion protein 6B11mGM as anti-idiotype vaccines a gainst ovarian carcinoma.
