摘要
目的探讨NF-κBp65在甲状腺癌中的表达及其与细胞周期的关系。方法应用免疫组化方法检测62例甲状腺癌、7例对照组织标本中p65蛋白的表达。用流式细胞仪分析甲状腺癌细胞周期分布。结果P65蛋白在正常甲状腺组织中未见表达,在甲状腺癌标本中的阳性率为62.9%(39/62),两者比较有显著性差异(P<0.05);乳头状癌、未分化癌、滤泡状癌和髓样癌p65阳性率分别为45.8%(11/24)、88.9%(8/9)、75.0%(12/16)和61.5%(8/13),各组之间比较有显著性差异(P<0.05);I期、II期、III期和IV期p65阳性率分别为41.2%(7/17),53.3%(8/15),77.3%(17/22)和87.5%(7/8),各组之间比较有显著学差异(P<0.05)。随着p65蛋白表达级别的增加,G0/G1细胞比例逐渐降低,而S期细胞比例及细胞增值指数PI均呈逐渐增高的趋势。结论NF-κBp65在甲状腺肿瘤组织中有异常高表达,其表达与甲状腺癌病理类型、临床分期及有无淋巴转移有关。P65蛋白表达强度与甲状腺癌细胞增殖活性相关。
Objective To detect the expression of p65 protein in thyroid cancer and to observe its clinical significance and its correlation with cell cycle.Methods Immunohistochemistry SABC method was used to examine the expression of p65 protein in sixty-two surgical specimens of thyroid cancer and seven control specimens ; Cell cycle was detected with Flow Cytometry.Results None of the seven normal control specimens was detected p65 positive expression and 62.9%(39/62) thyroid cancer specimens were detected p65 positive expression. P65 expression was significantly related with cell proliferation activity There were significant difference among different pathological type thyroid cancer and different clinic stage specimens.Conclusion The expression of p65 is associated with pathological type, clinic stage and cell proliferation activity of thyroid cancer.
