老年大鼠耳蜗酪氨酸羟化酶及单胺氧化酶-B基因的表达 被引量:2

The mRNA expressions of tyroxine hydroxylase and monoamine oxidase - B in the aged rat cochlea

摘要 目的 检测老年大鼠耳蜗多巴胺(dopamine,DA)合成和降解过程中的关键酶——酪氨酸羟化酶(tyroxinehydroxylase,TH)及单胺氧化酶-B(monoamine oxidase-B,MAO-B)基因的mRNA水平变化,以便更好地理解听觉系统老化的机制。方法 采用半定量逆转录聚合酶链反应(reverse transcription polymerase chain reaction,RTPCR)的方法检测耳蜗组织TH及MAO-B基因mRNA的含量。结果 耳蜗组织TH及MAO-B基因mRNAR的水平,老年组均高于成年组,差异均有非常显著性意义(tTH=9.634,tMA0-B=8.108,P<0.01)。结论 在基因转录水平证明了老年大鼠耳蜗多巴胺系统合成和降解过程发生了紊乱,推测耳蜗多巴胺代谢的变化在老年性聋的发生机制中可能起一定的作用。 Objective Dopamine is present in inner ear and play a role as an efferent neurotransmitter or neuromodulator. The objective of the present study was to understand the changes of dopamine metabolism in the aged rat cochlea. Methods Tyrox-ine hydroxylase and monoamine oxidase - B mRNA expressions were detected in the cochlea of the aged ahd adult rats by a semi -quantitative reverse transcription polymerase chain reaction method. Results Both tyroxine hydroxylase and monoamine oxidase -B mRNA levels in the cochlea of the aged animals were higher than that of the adult ones( tTH = 9.634, tMAO - B = 8.108, P< 0.01) . Conclusion The results showed that the process of synthesis and degradation of dopamine were disturbed in aged rat. Because of the important roles of dopaminergic innervation in cochlea, the metabolic alteration of dopamine would affect the modulation and protection of the primary afferent neurons and might play an important role in pathogenesis of presbycusis.
