摘要 目的 研究IFN- α对肿瘤细胞Colo205TRAIL功能受体DR4和DR5表达的调节作用,以及IFN -α刺激与肿瘤细胞对rTRAIL诱导凋亡敏感性的影响。方法 Colo205细胞以IFN- α刺激 0、24、48和 72h;采用RT PCR方法检测DR4和DR5基因表达;用间接免疫荧光染色结合流式细胞技术检测细胞表面DR4和DR5分子表达;采用annexinV和碘化丙锭双染色试剂盒检测细胞凋亡。结果 IFN -α刺激后,DR4和DR5的mR NA在细胞表面的表达均有增加,刺激 48h,膜型DR4和DR5表达达到高峰值 (16. 7%和 29. 8% );rTRAIL凋亡诱导效应在IFN- α刺激 48h的Colo205细胞中最高。结论 IFN -α上调死亡受体DR4、DR5的表达可能是其促进rTRAIL诱导肿瘤凋亡的重要机制。 AObjective Studying of regulating on IFN-α to the expression of DR4 and DR5, and its affection to rTRAIL-induced apoptosis in Colo205 tumor cells.Methods Colo205 cells were stimulated by IFN-α for indicated time period (0,24,48 and 72 hours),the expression of DR4 and DR5 gene were determined by RT-PCR assay.
