酸对食管鳞状细胞骨形态发生蛋白表达的影响

摘要 目的探讨酸对食管鳞状细胞骨形态发生蛋白4(BMP4)表达的影响。方法原代培养食管鳞状上皮细胞,采用不同pH值酸处理细胞,RT-PCR、实时荧光定量PCR与蛋白印迹分析方法测定BMP4、细胞内第二信使Smad1表达的变化,然后对其进行相关性分析。结果强酸刺激可明显增强食管鳞状上皮细胞表达BMP4、Smad1。结论酸可能通过促进BMP4及BMP信号通路相关蛋白表达而参与Barrett食管的发生。 Objective To explore the effect of acid on BMP4 expression in esophagus squamous cells.Methods Primary esophageal squamous cells were cultured,then the cells were treated with acid at different pH values.The changes of expressions of BMP4 and Smad1 were examined by RT-PCR,Quantitative real time RT-PCR and Western blot analysis.Results Acid at pH 4.0 obviously increased the expressions of BMP4 and Smad1 in esophageal squamous cells.Conclusion Acid can regulate the expressions of BMP4 and Smad1,which may play...
