摘要
目的优选附子水提醇沉工艺。方法应用正交设计安排实验因素,以酸性染料比色法测定总生物碱含量。比较并筛选不同工艺条件对总碱含量的影响。并对水提醇沉进行急性毒性试验和分离出的各部分进行镇痛实验。结果水提醇沉工艺的研究表明,水提液PH值对醇沉效果影响最大。小白鼠镇痛实验表明,醇沉沉淀物无镇痛作用,水提醇沉液有显著的镇痛作用。含等量总生物碱的附片液及附子水提液镇痛无显著性差异,且不显示毒性。结论本工艺能有效除去附子提取液中部分杂质,最佳工艺路线采用煮提液浓度0.75(生药)/ml,PH=5,80%的乙醇沉淀为最佳分离方案。
Objective To optimize the preparation process and determine the ideal conditions of Chuan-wu decoction by method of alcohol deposition. Method Orthogonal design was used to select test factors, and the acid dye colorimetric method was also used to determine the content of total alkaloids. The rats were used as an indicator of the abirritation with deposition and prepared Fu-Zi's decoction acute toxicity. Result The PH of Fu-zi decoction has a remarkable impact on the content of total alkaloids. No clear abirritation was found in treating rats by the deposition of alcohol, but the prepared decoction has a remarkable pain-alleviation effect and normal toxicity on rats. Conclusion The optimum preparation process is A2B3C3. The optimum preparation process is not only convenient for eliminating the impurity of Fu-zi decoction, also cost-effective and practicable.
