摘要
[目的]探讨四君子汤(Sijunzi Decoction,SJ)对小鼠H22肝癌细胞实验性肺转移的影响。[方法]建立小鼠实验性肺转移模型,用药后观察两个不同时间点肺表面转移结节的大小、数目及转移程度;ELISA法检测外周血中细胞间粘附分子-1(ICAM-1)、白介素-2(IL-2)的表达情况。[结果]四君子汤(SJ)能降低肺转移率,优于替加氟(FT-207)及生理盐水(NS)组。2周后SJ组、FT-207组肺转移结节数目分别为6.1±2.5和6.2±2.2,均低于NS组(7.3±3.5)(P<0.05);胸腺指数FT-207组较NS组、SJ组明显降低(P<0.01);血清中IL-2表达的量各组间无统计学意义(P>0.05),ICAM-1表达的量SJ组明显低于其他组(P<0.01)。4周后SJ组肺转移结节数目为6.3±2.7,明显低于NS组(7.8±3.2)和FT-207组(7.7±3.5)(P<0.01);脾脏指数FT-207组比其他组明显降低(P<0.01);血清中SJ组IL-2表达的量明显高于其他组(P<0.01),ICAM-1表达的量明显低于其他组(P<0.01)。[结论]四君子汤能抑制小鼠实验性肺转移,其机制可能是提高机体的免疫能力,调节肿瘤细胞黏附分子表达,避免肿瘤细胞从原发灶的脱落,并抑制肿瘤细胞与基质和内皮细胞的黏附,从而阻止肿瘤细胞的转移。
[Objective]To observe the effect of Sijunzi Decoction(SJ) on Hepatoma-22(H22) metastasis in mice.[Methods]The mouse experimental pulmonary metastatic model was established and then the mice were given Sijunzi Decoction by intragastric administration.After two various times the size and number and the aggravation grade of metastatic nodes on lung surface were observed;ELISA method was used to detect the expressions of ICAM-1and IL-2 in peripheral blood of mouse.[Results] Sijunzi Decoction Group was significantly better than other groups on decreasing the rate of metastatic focus.After two weeks,number of metastatic nodes on lung for SJ and FT-207 groups was 6.1±2.5 and 6.2±2.2 respectively,those were lower compared with NS Group(7.3±3.5);Thymus index numbers of FT-207 group were lower obviously than the other groups;expressed quantities of ICAM-1 in SJ group were much lower than the others.After four weeks,number of metastatic nodes on lung for SJ group was 6.3±2.7,that was lower obviously compared with group FT-207(7.7±3.5) and NS group(7.8±3.2);Spleen index numbers of FT-207 group was lower obviously than the other groups;To express quantities of IL-2 in serum,SJ group surpassed the others obviously,those of ICAM-1 were more lower than the others.[Conclusion] Sijunzi Decoction can inhibit the experimental pulmonary metastasis of mouse.The mechanism may be that Sijunzi Decoction may boost immune competence of organism and prevent the exuviations of tumor cells from primary carcinoma,inhibit the adhesion of tumor cells to extracellularmatrix(ECM) and endothelial cells,and to prohibit the metastasis of tumor cells.
