摘要
目的:研究生存素(survivin)反义寡核苷酸(antisense oligodeoxynucleotide,ASODN)对人乳腺癌MCF-7细胞株基质金属蛋白酶(matrix metalloproteinase,MMP)-2及MMP-9表达的影响,以进一步阐明生存素介导乳腺癌浸润及转移的机制。方法:经脂质体介导将生存素ASODN转染人乳腺癌细胞株MCF-7,用荧光显微镜检测转染效率,用逆转录聚合酶链反应(RT-PCR)、蛋白质印迹法检测转染细胞生存素、MMP-2及MMP-9表达改变。结果:荧光显微镜检测结果显示生存素ASODN可成功转染MCF-7细胞。与空脂质体组、空ASODN组相比,转染组细胞生存素、MMP-2、MMP-9 mRNA和蛋白的表达均显著降低。结论:生存素ASODN转染可有效抑制MCF-7细胞生存素基因的表达,并降低其MMP-2、MMP-9表达,可能对乳腺癌具有基因治疗的效果。
Objective: To investigate the effect of survivin antisense oligodeoxynucleotide(ASODN) transfection on the expression of matrix metalloproteinase(MMP)-2 and MMP-9 in human breast cancer line MCF-7 and to provide experimental foundation for survivin ASODN gene therapy for breast cancer.Methods: Survivin ASODN was transfected into MCF-7 cells through lipofectin.The rate of transfection was detected by fluorescence microscopy.The expressions of survivin,MMP-2 and MMP-9 in transfected cells were detected by RT-PCR and Western blot.Results: Survivin ASODN was successfully transfected into MCF-7 cells.After the transfection,RT-PCR and Western blot analysis revealed that the expressions of survivin,MMP-2 and MMP-9 mRNA and their proteins of antisense group were decreased significantly,compared with the control group.Conclusion: The transfection of survivin ASODN could effectively down-regulate the expressions of survivin,MMP-2 and MMP-9 in MCF-7 cell.Survivin ASODN may provide potential gene therapy for breast cancer.
