摘要 目的探讨4℃保存对人悬浮红细胞免疫功能的影响。方法采集健康献血者静脉血,制备悬浮红细胞,分别于保存前及4℃条件下保存1、2、3、4、5、7和9周时取样2 ml,采用直标法流式细胞术检测红细胞补体受体1(CR1)、白细胞分化抗原59(CD59)分子水平。结果红细胞CR1-GMFIR随4℃保存时间延长有逐渐增加的趋势,至4周时达最高值,之后逐渐下降,组间差异无统计学意义(P>0.05),与4℃条件保存前相比,除4周时CR1-GMFIR明显升高外,其他时间点的差异无统计学意义;而红细胞CD59-GMFIR呈逐渐下降趋势,组间差异有统计学意义(P<0.01),3~9周时CD59-GMFIR均明显低于4℃条件保存前。结论 4℃条件保存悬浮红细胞损害红细胞免疫功能,对红细胞CR1-GMFIR影响小,对CD59-GM-FIR影响大;保存5周内二指标受影响较小,保存2~3周时受影响最小。 Objective To study the effects of storing suspending erythrocytes at four centi-degrees on erythrocyte immune functions.Methods Suspending erythrocytes were prepared from venous blood drew from 13 healthy blood donors and stored at 4℃.Two milliliters of suspending erythrocytes was collected from 13 blood bags at pre-storage and 1,2,3,4,5,7 and 9 weeks after storage respectively,and complement receptor type 1(CR1) and cluster differentiation antigen 59(CD59) molecules on erythrocytes were measured by flow cytometry with direct fluorescent antibody technique.Results The erythrocytes CR1-GMFIR had an increasing tendency during storage of suspending erythrocytes at 4℃ from pre-storage to 4 weeks after storage and a decreasing tendency from 4 to 7 weeks after storage,but there were no significantly statistical differences in CR1-GMFIR among groups of different storage times(P>0.05).CD59-GMFIR of erythrocytes decreased gradually in 9 weeks,and there were statistically significant differences among groups(P<0.01),and CD59-GMFIRs at 3 to 9 weeks after storage were all lower than that at pre-storage(P<0.01 or P<0.05).Conclusions Storing suspending erythrocytes at 4℃ can impair erythrocyte immune functions,and CR1-GMFIR of suspending erythrocytes is slightly impaired,while CD59-GMFIR is severely affected.The impairment is slighter in 5 weeks and the slightest during 2~3 weeks.
