摘要
目的探讨高压氧环境下槲皮素在体外对肺腺癌A549/DDP细胞株耐药的逆转及其对P-gp和survivin蛋白表达的影响。方法 MTT法测定槲皮素在高压氧环境下对A549/DDP细胞耐药的逆转作用;流式细胞法(FCM)检测高压氧环境下A549/DDP细胞P-gp和survivin蛋白的表达。结果槲皮素在高压氧环境下使DDP对A549/DDP细胞的半抑制浓度(IC50)从(31.34±2.99)μg/ml下降至(7.56±0.87)μg/ml,其逆转倍数为4.15;高压氧处理A549/DDP细胞对P-gp的表达无显著影响(P>0.05),处理组AFI值为4.22±0.11,对照组AFI值为4.42±0.03;但能够显著降低survivin蛋白的表达(P<0.05),处理组AFI值为2.52±0.11,对照组AFI值为3.96±0.13。结论高压氧能增强槲皮素对肺腺癌A549/DDP细胞MDR的逆转作用;其增强作用可能通过降低survivin蛋白的表达实现。
Objective To evaluate in vitro effects of hyperbaric oxygen on reversal of quercetin (QUE ) to multidrug resistance of A549/DDP cells line,and the influence of hyperbaric oxygen to expression of P-gp and survivin protein.Methods Methyl thiazolyl tetrazolium ( MTT ) assay was used to determine the reversal effects of QUE in hyperbaric oxygen environment.Flow cytometry ( FCM ) was used to investigate the impacts on P-gp and survivin expression of A549/DDP cells treated by HBO.Results The 50% inhibitory concentration ( IC50 ) of DDP to A549/DDP cells reduced to ( 7.56±0.87 ) μg/ml from ( 31.34±2.99 ) μg/ml by the reversal effects of QUE and HBO,the reversal multiple was 4.15,which had significant difference.The levels of survivin protein expression in A549/DDP cells decreased significantly after interfered by HBO ( P<0.05 ),the AFI of HBO treated groups was 2.52±0.11,and control groups was 3.96±0.13; while there was no significant difference in the expression of P-gp protein in cells among the groups ( P>0.05 ),the AFI of HBO treated groups was 4.22±0.11,and control groups was 4.42±0.03.Conclusions HBO could enhance the effect of quercetin on reversal of MDR to A549/DDP cells,which might realize by decreasing the expression of survivin protein.
