摘要
目的研究脊髓缺血再灌注损伤时血清和脊髓组织中Ca2+、Mg2+、Cu2+、Zn2+的变化及其意义。方法 16只日本大白兔随机双盲分为两组(每组8只):假手术组和缺血再灌注组。缺血再灌注组采用夹闭肾动脉下腹主动脉40min造成脊髓缺血损伤。采用改良Tarlov评分评估手术后后肢神经功能和监测血清及脊髓匀浆中Ca2+、Mg2+、Cu2+、Zn2+的变化。结果 (1)同假手术组相比,缺血再灌注组出现显著的后肢神经功能受损(P<0.05);(2)缺血再灌注组缺血后血清Ca2+、Cu2+浓度逐步升高,再灌注后其浓度较再灌注前各时点值及假手术组对应值显著升高(P<0.05或<0.01),而缺血再灌注组再灌注后血清Mg2+、Zn2+浓度较再灌注前各时点值及假手术组对应值显著下降(P<0.05或<0.01);(3)术后第7天缺血再灌注组血清Ca2+、Mg2+、Cu2+、Zn2+浓度与脊髓匀浆的Ca2+、Mg2+、Cu2+、Zn2+浓度均分别具有正相关关系。结论脊髓缺血再灌注损伤时血清中Ca2+、Mg2+、Cu2+、Zn2+的浓度会出现相应的变化,监测血清中Ca2+、Mg2+、Cu2+、Zn2+的动态变化对监测有无脊髓缺血再灌注损伤有一定的指导意义。
Objective To validate the clinical significance and monitor the change of Ca2+,Mg2+,Cu2+ and Zn2+ in blood serum and spinal cord tissues during the spinal cord ischemic-reperfusion(I/R) injury in rabbits.Methods Sixteen white adult Japanese rabbits were randomly assigned to one of the 2 groups(8 rabbits per group) by the experimenters:GroupⅠ,namely,sham-operation group;GroupⅡ,namely,I/R group.Spinal cord ischemia was induced by infra-renal aortic cross-clamp for 40minutes in Group II.Post-operative neurological functions of back limbs and concentrations of Ca2+,Mg2+,Cu2+ and Zn2+ in the blood serum and the spinal cord tissues were assessed.Results Compared with the sham operation group,rabbits in the I/R group showed significantly worsened neurological functions as scored with the modified Tarlov criteria.With prolonged duration of I/R injury,serum Ca2+ and Cu2+ concentrations gradually increased,but Mg2+ and Zn2+ concentrations gradually decreased.Seven days after spinal cord injury,changes in Ca2+,Mg2+,Cu2+ and Zn2+ concentrations were significant.After 7 days of reperfusion,changes in the concentrations of Ca2+,Mg2+,Cu2+ and Zn2+ in spinal cord homogenates were consistent with those of the serum.Conclusions Significant changes in Ca2+,Mg2+,Cu2+ and Zn2+ concentrations in serum were noted during I/R injury.Monitoring the dynamic change of Ca2+,Mg2+,Cu2+ and Zn2+ in blood serum maybe can find the spinal cord I/R injury early in rabbits.
