诱导骨髓间充质干细胞分化为肉芽组织中血管内皮细胞的在体实验研究

摘要 目的:探讨骨髓间充质干细胞(MSCs)分化为肉芽组织中血管内皮细胞的可行性及其参与创面修复的可能机制。方法:抽取小型香猪的骨髓,经密度梯度离心分离、纯化间充质干细胞及培养扩增后,应用 BrdU 标记技术,以纤维蛋白胶为载体将已标记的细胞回植到提供骨髓的猪的全层皮肤缺损创面上,不同时间点(2、4、6、8、12周)取材,常规石蜡包埋、连续切片,行 BrdU 和血管内皮细胞第八因子(FⅧ)免疫组织化学染色,进行对比研究。结果:在细胞核内呈棕黄色着色的 BrdU 阳性细胞,多聚集在创面肉芽组织中的小血管周围,且有个别的血管内皮细胞也呈现 BrdU 阳性标记。同时 BrdU 阳性细胞胞浆中 FⅧ亦呈阳性表达。结论:创面愈合的过程中骨髓间充质干细胞与肉芽组织中小血管的形成有密切的关系;在创面微环境下骨髓间充质干细胞可能分化为血管内皮细胞并参与创面修复的过程。 Objective;To investigate the feasibility of differentiation of bone marrow mesenchymal stem cells (MSCs) into vessel endothelial cells,and to explore the potential relationship between this process and wound healing.Methods:Porcine MSCs were isolated from porcine marrow and grown in vitro.After labeling with BrdU, MSCs were engrafted to porcine full thickness skin wounds.At 2,4,6,8 and 12 weeks after the transplantation, immunohistochemical examinations were carried out to detect the positive staining of BrdU or Factor Ⅷ.Results: Most BrdU positive cells were aggregated around small blood vessels in the granulation tissues of the wounds.A few vessel endothelial cells,which expressed Factor Ⅷ,were also BrdU positive,and these cells were considered to be transplanted MSCs-derived cells.Conclusion:There was a close relationship between MSCs and the formation of small blood vessels in granulation tissues.Porcine MSCs have the potential to differentiate into vascular endo- thelial cells in injured skin.
