摘要
采用单因素和正交试验研究了氮源、蔗糖浓度和诱导子茉莉酸甲酯对蒙古黄芪愈伤组织生长和黄酮类物质积累的影响.结果表明:各因子影响黄酮积累量依次为:茉莉酸甲酯>蔗糖>水解酪蛋白>氮源;蒙古黄芪愈伤组织生长和黄酮含量积累的最佳培养基组合为:MS+NAA 1.0mg/L+6-BA 2.0mg/L+2,4-D 1.0mg/L+蔗糖50.0g/L+水解酪蛋白1.0g/L+MeJA0.02μmol/L,NH4+∶NO3-为1∶1.在这种条件下,黄酮产量达到最高为9.78mg/L.
To investigate the effects of nitrogen source,sugar concentration,elicitor methyl jasmonate on callus growth and flavonoids accumulation of Astragalus mongholicus Bge.The result indicated that the effectiveness of factors on Astragalus mongholicus Bge.callus growth and flavonoids accumulation was in this order:ratio of elicitor methyl jasmonate > sugar concentration > organic appendages hydrolysis of casein protein > nitrogen source,the best combination for callus growth and flavonoids accumulation of Astragalus mongholicus is MS + NAA 1.0 mg/L+ 6-BA 2.0 mg/L + 2,4-D 1.0 mg/L +source 50.0 g/L+hydrolysis of casein protein 1.0 g/L+ MeJA 0.02 μmol /L,NH4+∶NO3-is 1∶1.In these conditions,the highest flavonoid yield was 9.78mg / L.
