摘要
在室内人工条件下短期内增强UV B辐射(0 028~0 029J/(m2·s)),利用不连续非变性聚丙烯酰胺凝胶垂直电泳技术对3个不同地理种源的诸葛菜叶片过氧化物酶(POD)和细胞色素氧化酶(CYT)同工酶基因表达变化进行了研究.发现各个种源材料的POD和CYT酶Pod 2,Pod 3,Pod 4(重庆种源处理组除外)和Cyt-2位点基因表达与对照组相比均有明显增强,Pod 1和Cyt-1位点酶基因被激活而表达.并由于UV B辐射诱导产生许多新的同工酶带,这表明,短期内一定程度增强UV B辐射可以加强诸葛菜叶片POD和CYT同工酶的基因表达.
Under artificial conditions, changes of Peroxidase(POD) and Cytochrome oxidase(CYT) isozymes gene expression in leaves of three different geographic provenances of Orychophragmus violaceous(L.)O.E.schulz exposed to 0.028~0.029 J/(m^2·s) UV-B radiation were studied using the assay of vertical polyacrylamide gel electrophoresis(PAGE) in short term. Gene expression of some loci (including Pod-2, Pod-3, Pod-4and Cyt-2) of materials, from Beijing and Hangzhou under exposure to UV-B radiation were significantly enhanced by contrast to controlled groups. Only had gene expression of Pod-2, Pod-3 and Cyt-2 of material from Chongqing the same case as above. Being activated, gene of Pod-1 and Cyt-1 loci were expressed. In addition, inducement of UV-B radiation resulted in many new isozyme bands. All of above showed that enhanced UV-B radiation to some extent could strengthen gene expression on Peroxidase and Cytochrome oxidase osozymes in Orychophragmus violaceous (L.)O.E.schulz.
