摘要
目的研究Ebp1在膀胱癌组织中的表达情况,探讨Ebp1作用的分子机制和临床意义。方法通过应用免疫组化方法,检测Ebp1蛋白在52例膀胱癌组织、21例癌旁组织中的表达,并对Ebp1表达与膀胱癌临床病理指标的关系进行统计学分析。通过构建过表达载体,转染膀胱癌细胞株(T24),Westernblot检测Ebp1表达,对比观察转染细胞的增殖、细胞周期以了解Ebp1对肿瘤细胞的作用机制。结果临床大样本检测表明Ebp1蛋白在正常癌旁组织表达为强阳性(100%为阳性,且71.4%为强阳性),而在肿瘤组织中下调表达(63.5%≤弱阳性,P<0.01)。并且Ebp1蛋白的表达与肿瘤的TNM分期(P=0.003)、恶性程度分级呈负相关(P=0.001)。Ebp1过表达细胞株相比对照组生长较慢,流式检测表明G0/G1期细胞比例(73.2%)为对照组(28.43%)的2.5倍。结论 Ebp1通过细胞停留在G0/G1期而抑制细胞的生长,与膀胱癌的恶性相关,有望作为反映肿瘤恶性程度及预后判断的标记物和治疗靶标。
Objective To investigate the expression of Ebp1 in bladder cancer and the molecular mechanism of Ebp1 gene′s function,and discuss its diagnostic value in bladder cancer.Methods Immunohistochemical SP was employed to detect the expression of Ebp1 in 52 cases of bladder cancer and 21 cases of the adjacent-benign tissue to evaluate its clinical significance in the histological classification and prognosis of bladder cancer.T24 cell line with Ebp1 over-expression was setuped,cell proliferation,cell cycles and apoptosis were measured by CCK-8 kit and flow cytometry assay kit separately.Results The expression of Ebp1 decreased in bladder cancer compared the normal control(P<0.01)and was negative correlate with the cancer TNM stage(P=0.003)and malignant degree(P=0.001).Ebp1 over-expression cell grew slowly and most stay in G0/G1 phase compared with blank vector control.Conclusions Ebp1 plays an important role in the procession of bladder cancer and might acts as a valuable biomarker in monitoring on bladder cancer malignant degree and prognosis.
