摘要
利用RT-PCR和RACE技术克隆得到鲤鱼(Cyprinus carpio L.)X盒结合蛋白1(Xbp1)基因编码序列全长。生物信息学分析发现,鲤鱼Xbp1 mRNA的26 bp内含子及其两侧序列能形成典型的茎环结构,其蛋白结构具有1个保守的碱性亮氨酸拉链(bZIP)结构域;系统进化分析表明鲤鱼Xbp1与斑马鱼的亲缘关系最近;实时荧光定量PCR检测结果表明,两种亚型Xbp1 mRNA在鲤鱼不同组织中广泛表达,且未剪接型Xbp1 mRNA的表达量明显高于剪接型。
We cloned the full-length cDNA of X-box binding protein 1(Xbp1) in Cyprinus Carpio L.with RT-PCR and RACE.A conserved stem-loop structure formed by 26 bp intron and its flanking sequence in Xbp1 mRNA and a basic region leucine zipper(bZIP)domain in its protein structure are identified by bioinformation analysis.Phylogenetic analyses show that the carp Xbp1 has the closest relationship with zebrafish Xbp1.Real-time PCR analyses reveal ubiquitous expression of two subtypes of Xbp1 in different tissues of carp.Additionally,the main expression is unspliced Xbp1 mRNA rather than the spliced one under normal conditions.
