摘要
目的观察高强度聚焦超声对体外培养的胰腺癌细胞株Capan-1的生物学效应,分析高强度聚焦超声破坏肿瘤细胞的剂量与效应的关系。方法以高强度聚焦超声作用的胰腺癌Capan-1细胞株为研究对象,应用四甲基偶氮唑盐法测定细胞存活率,应用DNA梯度试验检测细胞凋亡,应用流式细胞术标记Annexin V&PI检测细胞凋亡。结果四甲基偶氮唑盐实验结果显示,随着高强度聚焦超声辐照强度和时间的增加,细胞存活率呈显著下降,其半致死量为269.8 W/cm2(7 s);DNA梯度试验与流式细胞术显示高强度聚焦超声具有诱导细胞凋亡的作用。结论高强度聚焦超声具有抑制胰腺癌Capan-1细胞生长的作用,同时具有诱导细胞凋亡的作用。
Objective To explore the biological effects with different irradiation times or intensity of high intensity focused ultrasound(HIFU) on pancreatic cancer cell line Capan-1 and investigate the relationship between the ultrasonic dosage and therapeutic effects.Methods Cancer cell line Capan-1 was used as the research object and the survival rate was detected by MTT assay in treatment with HIFU.The cell lines in treatment with HIFU were analyzed for apoptosis by DNA ladder and flow cytometer with Annexin V & PI staining.Results The results of MTT showed that HIFU significantly inhibited the growth of cell line Capan-1.With the increasing irradiation time and strength,the inhibitory effects increased significantly.The medial lethal dose values of HIFU on Cancer cell line Capan-1 was 269.8 W/cm2(7 s).The results by PCR and flow cytometer showed that irradiation of HIFU on Capan-1 cells induced cell apoptosis with a less dose.Conclusion HIFU could inhibit the growth of cell line Capan-1 and induce apoptosis cells.
