摘要
目的探讨葛根素防治激素性股骨头缺血性坏死的作用机制。方法大剂量地塞米松诱导体外培养的大鼠骨髓间充质干细胞成脂分化,在诱导成脂的同时给予不同浓度的葛根素进行干预。干预6 d后检测细胞内成脂标志物PPARγmRNA和C/EBPαmRNA的表达。结果葛根素干预组PPARγmRNA和C/EBPαmRNA的表达较对照组明显降低,差异具有统计学意义(P<0.01)。结论葛根素防治激素性股骨头缺血坏死的机理不仅是改善股骨头局部的微循环,同时还与其抑制激素诱导下的BMSCs成脂分化有关。
Objective To investigate the therapeutic mechanism of hormonal ischemic necrosis of femoral head treated by puerarin.Methods Rats bone marrow mesenchymal stem cells(BMSCs) were cultured in vitro and adipogenic differentiation of the cells was induced by high dose of dexamethasone,and different concentrations of Puerarin were added into the culture system simultaneously.After 6 days,the expressions of adipogenic markers,including PPARγ mRNA and C / EBPα mRNA,were detected.Results The expressions of PPARγ mRNA and C / EBPα mRNA in Puerarin groups were significantly lower than those of control group(P<0.01).Conclusion Puerarin has good effects on prevention and treatment of hormonal ischemic necrosis of femoral head,and its mechanism is not only related with the effect of promoting the microcirculation of femoral head,but also with the effect of inhibiting adipogenic differentiation of BMSCs induced by steroid.
