摘要
根据WRKY基因家族序列的保守区域设计简并引物,从甘薯(Ipomoea batatas L.)品种徐薯18中扩增出WRKY基因家族,经克隆、测序后得到15条WRKY基因家族序列,其编码的氨基酸序列包含有WRKY基因家族所具有保守的WRKYGQ和TTYEGKH(T/N/S/G/A/D)(H/Q)区。对甘薯WRKY基因家族氨基酸序列与部分植物WRKY基因家族氨基酸序列进行聚类分析,发现其聚为5类,相似性较高,推测各类中相似性较高的序列分别属于同一个基因家族。
The degenerate primers designed from the conserved motifs in the WRKY gene family were used to amplify WRKY gene family from Ipomoea batatas,Xushu18.15 WRKY gene family were obtained according to cloning and sequencing,and the amino acid sequence contained WRKYGQ and TTYEGKH(T / N / S / G / A / D)(H / Q) motifs of the WRKY.Cluster analysis of the predicted amino acid sequences grouped the WRKY of Ipomoea batatas and the known WRKY in NCBI database into five classes,and the sequence similarity was high.It was deduced that the RGAs with high similarity belonged to one gene family.
