丛枝菌根真菌孢子最适消毒方法及最佳培养基的研究 被引量:5

摘要 为了探讨离体培养条件下,丛枝菌根真菌(AMF)的最优萌发条件,试验以根内球囊霉菌(Glomus intraradices)为材料,研究了不同消毒方法以及不同培养基对根内球囊霉菌孢子萌发率的影响。结果表明:处理3(即采用无菌过滤膜过滤1%氯胺T+200 mg/L硫酸链霉素+100 mg/L硫酸庆大霉素+1%吐温20混合消毒液,同时延长消毒液的处理时间至15 min,消毒后用无菌蒸馏水漂洗6~10次)的灭菌效果最好,在避免杂菌污染的同时孢子萌发率可达80.67%;在4种培养基中,以琼脂培养基的效果最好,孢子萌发率最高,达80.67%;其次为琼脂蔗糖培养基,合成培养基和发根培养基的效果最差。因此,试验中的最适灭菌方法为处理3,最佳培养基为琼脂培养基。 In order to discuss the optimal germinating conditions of arbuscular mycorrhizal fungi(AMF) under in vitro culture, the influences of different disinfection methods and different culture media on germination rate of Glomus intraradices. The results indicated that, the treatment 3 had the best disinfection effect, which was using sterile filtration membrane to filtrate the mixed disinfection liquid(1% chloramine T + 200 mg/L of streptomycin + 100 mg/L of gentamicin+ 1% Tween 20) and extending disinfection time to 15 min, and then using sterile distilled water to rinse for 6~10 times,and the spore germination rate reached 80.67%. Among four kinds of culture medium, the agar medium had the best effect and the highest spore germination rate(80.67%), and the agar + sucrose medium had the second best effect, but the synthetic medium and the rooting medium had the worst effect. Therefore, the optimum disinfection method was treatment 3, and the optimal culture medium was agar medium.
