摘要
目的探讨骨桥蛋白(OPN)在宫颈癌组织及血清中的表达及与新辅助化疗敏感性关系。方法采用实时荧光定量PCR法检测49例宫颈癌患者(其中包括20例新辅助化疗患者),28例宫颈正常患者组织中OPN mRNA的表达及20例行新辅助化疗前、后宫颈癌组织中OPN mRNA的表达水平;同时采用ELISA法测定上述患者血清中OPN的表达水平。结果OPN在宫颈癌组织及血清中的表达均明显高于正常宫颈组织,且差异有显著性(P<0.05);新辅助化疗患者化疗后血清及组织中OPN的表达水平均明显下降(P<0.05)。结论OPN在宫颈癌中明显高表达,在宫颈癌的发生、发展中起着重要的作用,且与宫颈癌的新辅助化疗敏感性密切相关。
Objectives To investigate the expression of OPN in tissue and serum of cervical carcinomas,and the relationship with neoadjuvant chemotherapy sensitivity of cervical carcinoma.Methods Real-time quantitative reverse transcript ion-poly merase chain reaction(QRT-PCR) method was performed to detect the expression of OPN mRNA in28 cases of cervical normal patients and 49 cases of cervical carcinoma patients,including 20cases of neoadjuvant chemotherapy patients.At the same time,using ELISA method to detect the expression of OPN in serum of cervical carcinoma.Results The positive expression of OPN in tissue and serum of cervical cancers was significantly higher than in the normal patients,The expression levels of OPN in cervical cancer tissue and serum were significantly decreased after neoadjuvant chemotherapy(P<0.05).Conclusion The overexpression of OPN may he linked to the genesis and development of cervical carcinoma,and was significantly associated with neoadjuvant chemotherapy chemosensitivity of cervical carcinoma
