HSP90的功能及HSP90抑制剂的研究进展 被引量:6

摘要 热休克蛋白[1](heat shock protein,HSP)最初是在1962年Ritossa P研究果蝇唾液腺时发现的,是一种在热应激环境下合成并被激活的一组特殊蛋白质。随后研究进一步证明高温导致果蝇染色体出现蓬松是由于热休克使染色体内基因转录合成特异性的蛋白,因是在热应激条件下产生的新蛋白质,故称为热休克蛋白,又被称为应激蛋白。细胞在进行生长、发育、分化、基因转录等功能的时候,HSP发挥着重要作用。在应激状态下,保护细胞生命活动必需的蛋白质是其主要生物学功能,从而使细胞生存和生长得到维持。HSP需要与其他不同功能的多种蛋白形成复合体才能完成上述功能。HSP与靶蛋白结合或使其解离从而对靶蛋白活性功能进行调节,包括对靶蛋白的折叠、亚基之间的装配,并参与细胞内的运输及降解。在肿瘤细胞中多种HSP家族成员表达是增强的,说明HSP在肿瘤的发生及发展中具有重要作用。某些HSP在恶性肿瘤中常高表达,并保护肿瘤在不利环境中生长。HSP在生物体中是普遍存在的,它是一组高度保守的蛋白质,相对分子质量为(6~170)×103,是一个具有多成员的庞大的糖蛋白超基因家族。按照其相对分子质量大小可分为HSP110、HSP90、HSP70、HSP60、HSP (HSP25, HSP27,HSP28)五大家族。
