摘要
目的研究早期营养不良仔鼠海马中PI3Kp110α表达量的变化及其意义。方法将怀孕14 d的大鼠分为正常饮食组和半量饮食组,至哺乳期结束后仔鼠均正常饮食。测量仔鼠出生时体质量;胰岛素耐量试验(ITT)评估外周胰岛素敏感性;Morris水迷宫试验测试仔鼠15月龄时学习记忆能力;Western印迹、免疫组化(IHC)法分别检测脑组织PI3Kp110α及其信号下游分子Aβ1~42的表达。结果与对照组相比,实验组出生时体质量偏低(P【0.01);外周胰岛素敏感性降低(P【0.05);学习记忆能力下降(P【0.05);海马PI3Kp110α的表达量降低(P【0.05);脑组织Aβ1~42表达增加但差异无统计学意义(P】0.05)。结论早期营养不良对仔鼠老年期认知功能影响的机制可能与海马PI3Kp110α表达量异常有关。
Objective To investigate the expression of phosphatidylinositol-3 kinase catalytic subunit p110α( PI3Kp110α) in hippocampus of rats born with early malnutrition and explore the possible mechanism of the early malnutrition on cognition of elderly rats. Methods Fourteen-day pregnant rats were divided into control( normal diet group) and experimental groups( half amount of diet group). The newborn rats of the two groups were fed with normal diet after the postnatal 21 days. Birth weight was measured. The peripheral insulin sensitivity of rats was evaluated by insulin tolerance test. The ability of learning and memory of offspring rats were assayed by Morris water maze test. The expression of PI3Kp110α in hippocampus were detected by Western blot. The expression of Aβ1 ~ 42in cortex and hippocampus of offspring rats were detected by immunohistochemistry. Results with the control group,the mean birth weight of newborn rats were significantly lower( P<0. 01),the peripheral insulin sensitivity were lower( P<0. 05),the latency was longer( P<0. 05),the times of crossing platform was less( P<0. 05),the percent time in quadrant was significantly shorter( P<0. 01),the expression of PI3Kp110α in hippocampus was lower( P<0. 05) in experimental group,but the different expression of Aβ1 ~ 42 both in cortex and hippocampus were not statistically significant( P>0. 05). Conclusions Early malnutrition impairs the cognition of elderly rats,which may be related to the abnormal expression of PI3Kp110α in hippocampus.
