低分子量姬松茸多糖抑制Lovo细胞与内皮细胞黏附的作用及机制

摘要 目的观察低分子量姬松茸多糖(LMPAB)对人结肠癌Lovo细胞与内皮细胞黏黏附能力的影响及机制。方法免疫荧光法检测Lovo细胞与人脐静脉内皮细胞(HUVECs)黏附,流式细胞术检测Lovo细胞表面唾液酸化Lewisx(sLeX)表达。实时定量PCR检测Lovo细胞FUTⅦmRNA表达。结果 LMPAB减少Lovo细胞与内皮细胞的黏附,下调Lovo细胞FUTⅦmRNA表达,减少了sLeX的合成。结论 LMPAB具有抑制肿瘤细胞与内皮细胞黏附的作用,其机制可能与下调FUTⅦmRNA表达,减少sLeX合成有关。 Objective To investigate effect of low molecular weight polysaccharide( LMPAB) on the invasion ability of human colon carcinoma Lovo cells and HUVECs and its mechanism. Methods Lovo cells / HUVECs adhesion was assayed by Rose Bengal staining method. Lovo cells transmigrated monolayer HUVECs ability was examined by transwell chamber. The effect of LMPAB on sLeX in Lovo cell surface was detected by FCM. mRNA expression of FUTⅦ were determined by real-time RT-PCR. Results LMPAB could significantly decrease Lovo counts of adhesion to HUVECs. LMPAB could significantly inhibit Lovo counts that transmigrated monolayer HUVECs. LMPAB could significantly decrease sLeX on Lovo cell surface. LMPAB significantly down-regulated expression of FUTⅦmRNA. Conclusions By inhibiting tumor cells adhesion to endothelial cells,anti-tumor metastasis effects were complicated by LMPAB.
