固定酶催化合成十二烷基糖苷工艺试验与优化模型 被引量:2

摘要 为研制高纯度绿色环保型生物表面活性剂烷基糖苷(APG),通过单因素试验和二次正交旋转组合设计试验对ACA微胶囊固定化酶催化合成十二烷基糖苷工艺参数进行了优化,并对葡萄糖转化率模型进行了研究。结果表明:对葡萄糖转化率影响由大到小的因素依次为反应时间、加水量、反应温度、醇用量;反应温度与醇用量、加水量、反应时间,醇用量与加水量间的交互作用较大;最佳工艺参数如下:反应温度45℃、月桂醇8 mL、固定化酶5 g、葡萄糖1 g,不加水,反应时间为5天,葡萄糖转化率最大为97.9%。 In order to develop alkyl-polyglycoside(APG) which is a high purity and environment-friendly biosurfactant,the process parameters of APG synthesis catalyzed by ACA microcapsule immobilized enzyme were optimized through single factor experimentation and twice orthogonal rotating combination design test,and the model of glucose conversion rate was studied.The results show that factors which have influence on glucose conversion rate are: reaction time,amount of adding water,reaction temperature and amount of alcohol,what's more,the former is better than the latter;reaction temperature is more interactive with amount of alcohol,amount of adding water,reaction time,amount of alcohol and amount of adding water is the same;the best synthesis process parameters are as follows: reaction temperature at 45 ℃,amount of dodecanol of 8 mL,immobilized enzyme of 5 g,amount of glucose of 1 g,without adding water,reaction time of 5 days and the maximum of glucose conversion rate is 97.9%.
