摘要
运用限制性片断长度多态性技术(Restriction Fragment Length Polymorphism,RFLP)的衍生技术PCR-RFLP,构建我国赤潮藻的特征酶切图谱,达到对赤潮藻进行区分的目的。主要内容包括两部分:(1)运用PCR方法设计引物并扩增出了9种赤潮藻的18S rDNA片段。(2)选择了TaqⅠ和PshBⅠ为实验用酶,并对9种赤潮藻的核糖体18S rDNA片段进行RFLP分析,RFLP结果显示,对9种赤潮藻区分明显。
In this paper,the PCR-RFLP technology was used to construct the characteristic Macrorestriction Map of the red tide causing algae to distinguish them.The results comprise two aspects:(1) We acquired 18s rDNA segments of 9 species of red tide causing algae with the PCR technology.(2) TaqⅠand PshBⅠwere selected as the endonuclease in the research,and the 18S rDNA segments of 9 species of red tide causing algae were analyzed by RFLP.The results show that obvious distinction exists in 9 species of red tide causing algae.
