摘要
目的检测信号传导和转录激活因子-3(STAT3)、酪氨酸磷酸化STAT3(p-STAT3)及Survivin在胃癌及其癌前病变组织中的表达,探讨三者在胃癌组织中表达的关系及其临床病理意义。方法采用免疫组化二步法检测91例胃癌及其配对正常胃黏膜、肠化生及异型增生组织中STAT3和Survivin蛋白的表达,其中50例胃癌组织检测了p-STAT3的表达。结果STAT3、p-STAT3、Survivin蛋白在肠上皮化生(95.2%,71.4%,91.4%)、异型增生(85.7%,100%,100%)和胃癌组织(67.0%,60.0%,81.32%)中表达的阳性率均显著高于正常胃黏膜(6.6%,32%,3.9%),P<0.05;伴淋巴结转移胃癌组中STAT3、p-STAT3、Survivin蛋白表达的阳性率(67.7%,69.4%,86.8%)显著高于无淋巴结转移组(65.2%,35.7%,65.2%),P<0.05。在胃癌组织中STAT3与p-STAT3表达呈正相关(rk=0.288,P=0.013)。结论STAT3、p-STAT3及Survivin蛋白的过表达可能在胃癌的发生发展过程中起重要作用,是胃癌诊治中较有价值的标记物,但其具体分子病理学机制尚需进一步深入研究。
Objective To investigate the expression of signal transducer and activator of transcription 3(STAT3),tyrosine phosphorylation STAT3 (p-STAT3)and Survivin in gastric carcinoma and precancerous lesions,and to analyze their correlations with the clinicopathological features of gastric cancers. Methods PV9000 immunohistochemical method was used to detect the expression of STAT3 and Survivin in 91 gastric carcinomas,matched normal gastric mucosa,intestinal metaplasia and dysplasia. Fifty cases from 91 gastric carcinomas mentioned above were also evaluated for the expression of p-STAT3. Results The positive rates of STAT3,p-STAT3 and Survivin expression were significantly higher in intestinal metaplasia(95.2%,71.4%,91.4%),dysplasia(85.7%,100%,100%)and gastric carcinoma(67.0%,60.0%, 81.32%)than in normal gastric mucosa (6.6%,32%,3.9%),P < 0.05. In gastric cancer with lymph nodes metastasis,the positive rates of STAT3,p-STAT3 and Survivin protein(67.7%,69.4%,86.8%)were significantly higher than that in the group without lymph nodes metastasis (65.2%,35.7%,65.2%),P < 0.05. In gastric carcinoma STAT3 expression was positively correlated with p-STAT3 expression(rk =0.288,P =0.013). Conclusion The up-regulation of STAT3,p-STAT3 and Survivin may play important roles in the occurrence and progression of gastric cancer. STAT3,p-STAT3 and Survivin may be markers of value in gastric cancer diagnosis and treatment,but the specific molecular pathological mechanism still require further investigation.
