摘要 目的 :确定鸡胚尿囊膜模型血管生成指标 .方法 :3 0只发育良好的 1 0日龄鸡胚分为 3组 (每组 1 0只 ) :rbFGF,kringle 5和NS组 .分别吸取rbFGF ,kringle5 ,NS液 1 0 μL于甲基纤维素膜上 ,加于鸡胚CAM上 .用计算机图像分析系统对鸡胚CAM血管生成的血管数目 (VN)、血管平均管径 (VD)、血管面积 (VA)、血管面积 /鸡胚CAM面积之比值 (VA/CAM)四项指标进行定量 .结果 :Kringle 5组的VN ,VA ,VA/CAM值显著低于生理盐水 (NS)对照组 (P <0 0 5 ) .rbFGF组的VN ,VA ,VA/CAM值显著高于NS对照组 (P <0 0 5 ) ,各组间VD无显著差异 (P >0 0 5 ) .血管定量指标VN和VA ,VA/CAM值之间的相关系数分别为 :0 967,0 973 (P <0 0 1 ) ;回归系数分别为0 947,0 93 5 (P <0 0 1 ) .结论 :与常规定量指标VA相比 ,VA/CAM值更为客观有效 . Objective:To study the quantitative indexes of the chick embryo chorioallantoic membrane(CAM)in angiogenesis models.Methods:Thirty well-developed eggs were divided into three groups of ten for each group:rbFGF,kringle 5 and NS group(n=10),respectively subtracted 10 μL of each onto methylcellulose discs and added into CAM.Computer image analysis system(CIAS)was used to quantify angiogenesis with the vascular number(VN),vascular diameter(VD),vascular area(VA),the ratio between the vascular area and the CAM area(VA/CAM).Results:VN,VA and VA/CAM index of Kringle 5 were lower than that in the control group(P<0.05),and those of rbFGF were higher than that in the control group(P<0.05)with no big difference in VD(P>0.05).The correlation coefficients of VN between VA and VA/CAM were respectively 0.967 and 0.973(P<0.01);the regression coefficients were respectively 0.947 and 0.935(P<0.01).Conclusions:Among several evaluating indexes of angiogenesis,VA/CAM is more effective and objective.
