摘要 背景与目的:胶质瘤的放疗效果与放射敏感性密切相关,有许多因素影响着胶质瘤的放射敏感性,其中胶质瘤的分子特征是决定放射敏感性的重要因素之一。本研究探讨了不同的基因表达与胶质瘤放射敏感性的关系。方法:采用X线照射及集落形成试验检测7种人脑胶质瘤细胞株的存活分数SF2;采用Western blot法检测细胞株中Bcl-2、p53、p21、MAPK、Ras、p-p53(磷酸化p53)的蛋白表达水平;分析基因表达与放射敏感性的相关关系。结果:7种不同胶质瘤细胞株的存活分数显著不同,从0.19到0.77,平均值为0.46;Ras、Bcl-2和MAPK表达与胶质瘤的放射敏感性有明显的相关,相关系数分别为0.81、0.83、0.70;而p53、p-p53、p21则与放射敏感性无明显相关,相关系数为0.21、0.17、0.17。结论:胶质瘤细胞存在不同放射敏感性的细胞群,这些细胞的敏感性与某些基因的表达有密切关系。Ras、Bcl-2和MAPK的表达与胶质瘤的放射抗拒性相关,而p53、p-p53、p21的表达与胶质瘤的放射敏感性关系不明显。 BACKGROUND & OBJECTIVE: The efficacy of radiotherapy for glioma is influenced by the radiosensitivity, which is affected by many factors. The molecular characteristic is one of the most important factors determining radiosensitivity. This study has analyzed the radiosensitivity of glioma cell lines and related gene expression. METHODS: X-ray (2Gy of irradiation) and colony forming experiment were used to detect surviving fraction(SF2) for 7 glioma cell lines. The expression level of Bel-2, p53, p21, MAPK, Ras, p-p53 were determined by using western blot analysis. Then, the correlation between gene expression and radiosensitivity was analyzed. RESULTS: The SF2 in 7 glioma cell lines varied between 0. 19 and 0.77 (average 0.46). The expression of Ras、Bel-2 and MAPK was significantly associated with the radiosensitivity of glioma cell lines with the correlation coefficient of 0.81, 0.83, and 0.70, respectively. However, the expression of p53, p-p53 and p21 showed no significant correlation with the radiosensitivity (the correlation coefficient was 0.21, 0.17, and 0.17, respectively). CONCLUSION: Human glioma cell lines possess different radiosensitivities which could be determined by the expression of certain genes. The expression of Ras, Bel-2 and MAPK correlates with radiosensitivity, while p53, p-p53 and p21 do not associate with the radiosensitivity of glioma.
