摘要
目的 观察降钙素基因相关肽(CGRP)对内皮细胞增殖及内皮功能的影响。方法 检测了培养内皮细胞周期时相的分布以及细胞内游离钙离子的水平,同时对培养液中一氧化氮(NO) 和内皮素(ET) 的含量进行了测定。结果 加用CGRP组,S期细胞明显最多,细胞内游离钙离子水平增高,培养液中NO含量升高,ET含量降低( P均< 0-01)。结论 CGRP促进血管内皮细胞增殖,其作用可能与增加细胞内钙离子水平有关,此外,它还能促进NO 的合成,降低ET的水平,因此对改善内皮细胞的功能,防治动脉粥样硬化起着积极的作用。
Objective To observe the effects of CGRPon cultured endothelial cell.Methods The effect of CGRPon cell cycle kinetics of cultured endothelial cells wasinvestigated byflow cytometry.In addition ,the cytoplasmicfree calcium turnover was also tested . Nitric oxide (NO) and endothelial (ET) ,two compounds re leased by endothelialcell,weretestedin orderto observe the effect of CGRPon secretion of endothelial cells.Re sults As compared with controlgroups,CGRPincreased Sphase percentage of endothelialcells,thelevelofcyto plasmicfree Ca2 + ,the NOlevel,decreased ETlevel of the conditioned medium .Conclusion CGRPstimulated proliferation ofand endothelialcellsincreasedthelevelof cytoplasmicfree Ca2 + indicating an involvement ofcycto plasmicfree Ca2 + assecond messagerin thisreaction.In addition,CGRPpromoted endothelial cellsto secrete NO and also inhibit ET.
