非小细胞肺癌中检测肿瘤新生血管的方法学探讨

摘要 为了探讨Ⅷ因子相关抗原(F8RA/vWF)显示肿瘤新生血管的可靠性及方法学。我们对85例非小细胞肺癌(nonsmalcellungcarcinoma,NSCLC)的石蜡组织切片进行F8RA/vWF免疫组化染色,结果显示:F8RA/vWF免疫组化清楚地选择性地显示血管内皮细胞,肿瘤细胞和其它正常组织不着色,肿瘤内有高密度微血管区。微血管数目均值为71±39,Ⅰ期NSCLC微血管数目均数为652±348,腺癌为766±392。本结果与其他研究结果比较:F8RA/vWF免疫组化显示肿瘤新生血管有一定的可靠性。 In order to study the reliability and methodology of factor Ⅷ related antigen/von Willebrands factor(F8RA/vWF) staining in the detection of neovascularization of tumor, paraffin tissue sections were stained for F8RA/vWF by a standard immunohistochemistry technique in 85 cases of non small cell lung carcinomas (NSCLC). The results showed that F8RA/vWF positive staining provided reliable discrimiration of endothelial cells. Tumor cells, as well as other normal tissue constituents, were consistently unstained. There were the highest neovascularization areas in intratumor region. The mean microvessel counts, mean microvessel counts in stage Ⅰ and mean micovessel counts in adenocarcinoma were 71±3 9、65 2±34 8 and 76 6±39 2 respectively. Comparing our results with others, it is concluded that the F8RA/vWF staining has been shown to be reliable to evaluate the neovascularization of tumors. A standard immunohistochemistry technique and reproducible method of microvesel measurement are needed.
