摘要
肿瘤坏死因子（ＴＮＦ）对甲状腺细胞生长增殖有何影响尚未定论，作者采用四氮唑蓝（ＭＴＴ）比色法测定ＴＮＦ－α对鼠ＦＲＴＬ－５细胞生长增殖的影响。结果提示：在不存在ＴＳＨ条件下，ＴＮＦ－α在２００～２００００ｋＵ／Ｌ范围内对ＦＲＴＬ－５细胞既无明显细胞溶解作用，亦无明显促增殖作用；在ＴＳＨ存在条件下，上述剂量ＴＮＦ－α对ＦＲＴＬ－５细胞增殖有轻～中度抑制作用。ＴＮＦ－α对甲状腺细胞增殖的抑制作用，可能不是通过其细胞毒作用，而主要是通过抑制ＴＳＨ作用来实现。
Whether tumor necrosis factor-α(TNF-α)influences the proliferation of rat FRTL5cells has not been certified.We used tetrazolium salt(MTT)colorimetric assay to study the effects of TNF-αon the proliferation of FRTL-5cells.When the cells were cultured in 5H(TSH-free)medium,neither proliferative responses of FRTL-5 cells induced by TNF-α(from 200 kU/L to 20 000 kU/L)were found,nor were the cytolytic effects of TNF-αHowever,in 6H medium(containing TSH),the proliferative responses of cells to TSH were moderately inhibited by TNF-αin the levels from 200 kU/L to 30 000 kU/L,and there were no differences between the different dose groups.It suggested that the inhibitory effects of TNF-αmight be due to inhibition of the responses of FRTL5 cells to TSH.
