摘要 用核酸分子杂交技术研究多胺生物合成限速酶L-鸟氨酸脱羧酶(ODC)的不可逆抑制剂—二氟甲基鸟氨酸(DFMO)对HL-60细胞c-myc、c-fos、c-Ha-ras癌基因及ODC基因表达的影响。结果表明DFMO处理的HL-60细胞中,c-myc癌基因表达减少,c-fos基因表达水平上升,而DFMO对ODC基因表达无明显影响。此外,本文证实了加入外源性腐胺能阻止DFMO引起的上述变化,表明DFMO对HL-60细胞基因表达的影响是其抑制细胞内多胺生物合成所致。 The effects of α-difluoromethylornithine (DFMO) on the expression of c-myc, c-fos, c-Ha-ras oncogenes and ODC gene of HL-60 cells were investigated. Analysis of dot blot and Northern blot indicated that a decrement in the expression of c-myc and c-Ha-ras oncogenes and an increase in the c-fos expression occurred in HL-60 cells treated with DFMO. No changes had b(?)n observed in the expression of ODC gene of the cells treated with DFMO. In addition, it was observed that the changes in oncogene expression resulted from DFMO treatment can be prevented by the provision of exogenous putrescine, indicating that the effects of DFMO are due to interference with the polyamine biosynthesis of HL-60 cells.
