摘要
0.02、0.04、0.06mmol ASI和0.008、0.016、0.024mmol SK可对抗远志皂苷的溶血作用,且随剂量的增加而增大。12.5mg/kg ASI及6mg/kg SK,ig每日二次,连续5天,可对抗CCl_4所致死亡。12.5、25mg/kg×d ASI,6、12mg/kg×d SK,ig持续14~16天,可引起昆明种小鼠或C_(57)BL/6小鼠的红细胞内超化歧化酶活性明显增加。12.0mg/kg×d SK、25mg/kg×d ASI持续4天,可使昆明种小鼠或C_(57)小鼠的肝内MDA含量明显下降,GSH含量明显上升。以上结果提示黄芪苷具有抗生物氧化性质,这不仅可能有助于解释黄芪苷对抗CCl_4的毒性作用和远志皂苷的溶血作用,这一基本生化机制,也有助于阐明中医应用黄芪异病同治的原理。
Astragalus Saponin I(ASI), Astrasieversianin Ⅺ(SK)were separately isolated by C-hemistry Dept, in our College and Shanghai Institute of Chemistry cooperated with Xinjiang Institute of Chemistry. It was found that Tenuigenin was known to facilitate he-molysis. The effect of its hemolysis was tested in a different incubation time ( 2, 4 , 24 h) in the presence of ASI (0.02, 0.04, 0.06 mol) , SK(0.008, 0.016, 0.024mol) resulting in a marked decrease in Hb which was dependent on the concentration of ASI and SK, 85 male mice were respectively given 12.5, 25 mg/kg.d ASI and 6 , 12 mg/kg.d SK for 14-16 days resulting in significant increase in erythrocyte superoxide dismutase activity(SOD) and dose dependent.Its activity rose from 5135+834 to 9785+464 maximum in Kun Ming species and from 4326+965 to 6336+684 (X+SD, u/g Hb)in C57EL/6 species. 100 mice were treated with ASI 12.5mg/kg, SK 6mg/kg or 1 % CMC 10 ml/kg ig for 5 days,then each mouse was intraperitoneally injected a dose of 40% CCl4 6 ml/kg. The total number of mice pretreated with ASI or SK group and died by the toxicant was lower than that of control group in five days.Mice were grouped and given different dosage for 5 days and shown no significant increase in protein content, but significant decrease in MDA and significant increae in GSH.It suggested that ASI and SK had a antioxidative effect since she contents of GSH and SOD were enhanced and MDA was decreased by them. The basic mechenism of the action may be an important bearing upon different diseases rendered with equal Huangqi in Chinese traditional medicine theory.
