摘要
目的对微血栓法制作大鼠痴呆模型的方法进行改良,以提高成模率。方法选取20只Wistar大鼠,经大鼠颈外动脉逆行穿刺到颈总动脉与颈内动脉交口处注入微栓子,制作多发性梗塞痴呆模型。结果 20只参与造模的大鼠中,1只在造模过程中死亡,2只偏瘫过于严重,2只在术后两天死亡。剩余15只大鼠采用Morris水迷宫评价其认知能力,其中12只存在认知功能减退。结论改良方法可减少大鼠在造模术中的死亡,提高成模率。
Objective The method of homologous embolus induced multi-infarct dementia in rats was improved to enhance the success rate of operation. Methods Twenty Wistar rats were used in this study. The rat model of multi-infarct dementia was established. The external carotid artery of the rat was exposed and the homologous embolus was injected retrograde through the external carotid artery into the intersection between the internal carotid artery and common carotid artery. Results One of twenty rats dead in the modeling process,two suffered serious hemiplegia,and two dead two days after the operation. The other rats were given Morris water maze test to evaluate the cognitive abilities. Results of Morris water maze test showed cognitive impairment in twelve of fifteen rats. Conclusion The improved method could reduce the death rate and increase the success rate in the rat model.
