摘要
为了研究神经肽Y(Neuropeptide Y,NPY)对肠道上皮细胞完整性及通透性的影响,本试验采取不同浓度和不同处理时间的TNF-α诱导的Caco-2细胞凋亡为模型,研究了不同浓度NPY对TNF-α诱导的Caco-2细胞凋亡的影响。选择处于对数生长期的Caco-2细胞,将试验组分为对照组、TNF-α组、NPY组、NPY+TNF-α组。采用MTT法检测NPY、TNF-α的细胞毒作用,利用流式细胞技术检测NPY对TNF-α诱导的Caco-2细胞凋亡的影响。结果显示,10nmol/L的NPY对Caco-2无细胞毒作用;与NT组相比,30μg/L TNF-α刺激24h后显著引起Caco-2的凋亡;与TNF-α组相比,NPY显著抑制TNF-α诱导的Caco-2的凋亡。结果表明,10nmol/L的NPY对TNF-α诱导的Caco-2凋亡有保护作用。
In this experiment,in order to investigate the effective mechanism of NPY on TNF-α-induced Caco-2cell apoptosis,Caco-2cells,in the logarithinic growth phase,were treated with TNF-αand/or NPY.MTT assay was used to detect the cytotoxicity of NPY and TNF-α,flow cytometry was used to explore the impact of NPY on TNF-α-induced Caco-2cell apoptosis.The results showed that there are no cytotoxicity on Caco-2cells under 10nmol/L NPY treatment;compared with the NT group,30μg/L TNF-αafter 24 hstimulation induced apoptosis significantly in Caco-2cells;10nmol/L of NPY significantly inhibited TNF-αinduced Caco-2cells apoptosis.The results indicated that NPY has a protective effect on Caco-2cells apoptosis induced by TNF-α.
