芦荟凝胶促进糖尿病创面溃疡愈合的机制 被引量:8

摘要 目的在分子水平探讨芦荟凝胶促进糖尿病创面溃疡愈合的机制。方法实验分为3组,A组为正常组,B组和C组应用链脲佐菌素诱导的糖尿病大鼠模型并建立全皮伤口模型,B组给予生理盐水处理创面,C组给予芦荟凝胶干预,4次/d,通过胶原纤维-Masson三色法和免疫组织化学法评估伤口形成后3、7、14 d皮肤组织的组织病理学改变,采用逆转录-聚合酶链反应(RT-PCR)检测大鼠皮肤基质金属蛋白酶(MMPs)-9和金属蛋白酶抑制剂(TIMPs)-1 mRNA的表达情况。结果 B组大鼠伤口愈合明显迟缓。芦荟凝胶干预可加速糖尿病大鼠大鼠伤口愈合。伤口愈合过程中,B组MMP-9 mRNA水平持续高于A组(P<0.01),而TIMP-1 mRNA水平持续低高于A组(P<0.01);给予芦荟凝胶干预的C组MMP-9 mRNA和TIMP-1 mRNA水平仅在伤口形成第3天与A组相比具有统计学差异,其余时间点均与A组相近。芦荟凝胶干预可降低大鼠伤口组织MMP-9 mRNA/TIMP-1 mRNA比值。结论糖尿病大鼠皮肤伤口愈合速度明显减慢,芦荟凝胶干预可加速糖尿病大鼠伤口愈合,芦荟凝胶的这种作用可能是通过调控MMP-9/和TIMP-l水平实现的。
